Loading raw Ct data
Once satisfied that the file has been loaded click the banner for section 3 and proceed to the design modification section.
Specify the file in PowerNest using the ‘Select file’ button to the left of the table. In order that the data be recognised by PowerNest they must be presented in a specific format (an example of which can be downloaded using the link to the left).
NB: The blue column- and row-headers must remain intact as powerNest uses these to identify the location of the data within the input file.
PowerNest will read raw Ct data from a file and provides a fast and intuitive method for grouping the data into the correct position in the experiment design hierarchy.
Once the selected file has been successfully loaded the table in powerNest will be populated with Ct data. It is now necessary to provide information on the structure of the assay from which these data were produced so that powerNest can perform the statistical analyses required to produce error estimates for the processing levels. This is achieved by selecting cells from the table with the mouse (individually, or using the shift-key for multiple cell selection) and dragging them onto an RT node in the tree below.
On recognition of a successful allocation, the RT node to which the data were dropped will turn green, as will the cells in the table above containing the dropped data.
The allocation of data for any given node in the experiment hierarchy can be checked at any point by simply selecting it with the mouse. Clicking a node, at any level, will highlight the cells in the table above that contain the data allocated to that node and its sub-nodes. To remove an allocation, right-click the desired node and select ‘Remove allocated Ct data from this node’ from the pop-up menu.
